      经过比较研究,虫媒病毒单扩散溶血试验的适宜条件为:每0.1ml压积绵羊红血球需0.25ml蔗糖丙酮法制备的乳鼠脑抗原致敏;于28℃孵育优于37℃,绵羊红血球在此两种温度下均发生特异溶血,而鹅红血球仅在37℃能溶血,28℃下结果均为阴性。单扩散溶血试验的敏感性相当于血凝抑制试验。用C6/36细胞培养的病毒作抗原,首次作成功了Chikungunya、Sindbis,乙型脑炎和Kunjin单扩散溶血试验,其特异性和敏感性与乳鼠脑抗原相近。用21种虫媒病毒小鼠免疫腹水与14种虫媒病毒乳鼠脑抗原做了交叉单扩散溶血试验。

      Through comparative studies, the optimal conditions for single radial haemolysis (SRH) for the detection of antibodies to arboviruses are as follows:(1) 0.1ml packed sheep red blood cells (SRBC) which were sensitized with 0.25ml sucrose-acetone suckling mouse brain antigens gave larger specific haemolytic zones than that sensitized with 0.4ml antigen. (2) The incubation temperature at 28℃ was better than that at 37℃. Sensitized SRBC incorporated in gel developed specific haemolytic zones at both temperatures.whereas sensitized goose cells developed zones only at 37℃.Furthermore,culture fluid of C6/36 cells infected by virus was successfully used for SRH instead of using sucrose-acetone antigens,so that the procedures were largerly simplified. The culture fluids of C6/36 cells infected by Japanese encephalitis (JEV), Kunjin, Chikungunya and Sindbis viruses gave results as sensitive as those given by sucroseacetone antigens. Cross SRH was performed with 21 antiarbovirus ascitic fluids and 14 arboviruses antigens.It was found that there was no cross reaction among three group A viruses, between group A and B arboviruses, and between mosquito and tick-boren flaviviruses, whereas close relationships were observed among four JEV subgroup viruses anor tick-borne encephalitis complex. The extent of cross reactivity in SRH was lower than haemagglutination inhibition test and somewhat higher than complement fixation test.Having such a characteristic, SRH could be applied to the identification of newly isolated arboviruses.
