      目的从基因组全序列分析探讨浙江省1 998--2009年H3N2亚型流感病毒的基因变异与流感流行的关系.方法选择浙江省1998--2009年流感流行期间分离到的H3N2亚型流感病毒流行代表株19株,采用RT-PCR方法扩增其8个基因片段后进行序列测定,并与近十余年间使用的疫苗株序列进行比较.结果H3N2亚型流感流行株HA和NA基因的氨基酸变异率最高,分别为13.98%和10.00%;其他6个内部基因所表达的蛋白中,除NP、M2和NSl(氨基酸变异率分别为6.43%、6.19%和3.48%)外,其他蛋白的氨基酸变异均<3.00%;在H3N2亚型流感存在较大范围流行的年份,除HA和NA基因外,流感病毒其他6个内部基因也会发生不同程度的变异.此外,在以往一些年份中,WHO推荐的H3N2流感病毒疫苗株与浙江省同期流行株之间,在许多基因上存在着滞后现象.结论在加强对H3N2亚型流感毒株表面HA与NA基因监测的同时,也应注意到其他内部基冈的变异,为新一轮流感流行株的出现提供重要的预警信息.

      Objective To analyze the relationship between influenza epidemic and genetic characteristic on the whole genome of influenza virus subtype A,H3N2 strains isolated in Zhejiang province during 1998 tO 2009.Methods All of the eight genes from the 19 Zhejiang influenza virus isolates.circulated during 1998 to 2009.were amplified by RT-PCR and sequenced.The obtained sequences Wfl'e aligned and analyzed with the vaccine strains being used in the Iast 10 years. Results The highest mutation happened within HA and NA genes and the amino acid divergent ratios were 13.98%and 10.00%.Amongst the six intemal proteins.the amino acid divergent ratios of NP.M2 and NSl were 6.43%,6.19%and 3.48%respectively,and the others were lower than 3%. Other than the HA and NA genes。mutations were also observed on six internal genes of the strains isolated in those years when the influenza virus subtype A/H3N2 was widely circulating. Additionally,there had been all obvious genetic lag between vaccine strains recommended by WHO and the contemporary Zhejiang epidemic strains for many years.Conclusion Besides on HA and NA genes.surveillance programs should also be covered mutations regarding the internal genes of influenza virus subtype A/H3N2 strains.in order tO provide important information for forecasting and warning of a new round of influenza epidemic.
