| 目的 研究宁夏地区手足口病患者中肠道病毒71型(EV71)分离株的基因特征。方法 采集手足口病患者标本进行病毒分离培养, 阳性标本进行RT-PCR鉴定后, 选取29株EV71分离毒株, 对其VPI区进行核苷酸序列测定, 并参考A、B、c各基因型的参考毒株和以往中国EV71的分离毒株进行同源性分析并构建系统发生树。结果 从439份标本中分离肠道病毒215株, 经RT-PCR鉴定, 93株为EV71。选取其中29株进行VPl基因全序列测定, 根据VPl全基闲序列构建系统发生树, 29株毒株均在c基因型中, 与c4亚型属同一分支, 与G4亚型代表株核苷酸同源性为91.7％～99.4％, 氨基酸同源性为96.6％一100.0％。结论 宁夏地区手足口病患者中分离的EV71毒株属c基因型的c4亚型。 |

| Objective To study the genetic characterization of enterovims 71(EV71)strains isolated from specimens of patients with hand-foot-mouth disease(HFMD)in Ningxia province in 2008. Methods AII the St001.throat swab and vesicle samples that collected from patients with HFMD were cultured.The positive isolates were identified by reverse transcriptase PCR(R, r-PCR)with specific primers of EV71.Complete VP l gene sequences(89 l nucleotides)of 29 strains(part of 93 EV7l strains) were determined and compared with A.B and C genotype reference EV71 strains while EV7l China isolates by homogeneity and phylogenetic tree analyses.Results 2l5 strains of EV were isolated from 439 specimens.Results from PCR indicated that 93 strains belong to EV71.Phylogenetic tree analysis revealed that the selected 29 stains were clustered with reference strains of subgenotype.The nucleotide identity C4 reference strains was 91.7%-99.4%.The amino acid homogeneity was 96.6%-l 00.0%.Conclusion The recently identified EV7l strains in Ningxia province belonged to subgenotype C4 which resembled to most of the isolates in China. |
