Abstract:
      本文报道了ELISA定量检测血清白喉毒素抗体的方法。经测定43名儿童血清ELISA滴度与锡克氏试验结果比较,锡克氏试验阳性与阴性血清的ELISA平均滴度分别为1:82.0及1:219.9,以ELISA滴度 ≥ 1:100及<1:100表示有或无免疫力,则与锡克氏试验的符合率为70%。我们对粗制毒素进行了纯化,用以作为抗原可进一步提高试验的敏感性。实验证明ELISA的特异性、敏感性和重复性均好,而且只需微量(10微升)血清,操作简便、为流行病学调查提供一个新的定量检测方法,可用以替代锡克氏试验。
English Abstract:
      This paper gives the result of quantitative estimation of antibodies to Diphtheria toxin in sera by ELISA method. The titers found with ELISA was compared with the results of Shick's test in 43 children.The average ELISA titers for Shick's positive and negative sera were 1:82.0 and 1:219.9 respectively. We considered that a titer ≥ 1:100 by ELISA representing an efficient immunity and an ELISA titer<1:100 representing an defficient immunity. Consequently a correspondence with the result of Shick test being 70%.When the crude diphther a toxin was purified and used as antigen in ELISA, there was an increase in sensitivity with this method. ELISA was found to be specific, sensitive and reproducible and the amount of sera required was only 10ul.The advantage of using this method is both simple and easy.It serves a better method of immunity in epidemiological survey and could take the place of Shick's test.
