Abstract:
      感染恶性疟原虫的夜猴血球经超声处理、低温高速离心后可获得高效价的恶性疟原虫可溶性抗原。该抗原吸附于聚苯乙烯塑料板上于室温下可保存抗原性一年以上,适于现场试验及流行病学调查。在ELISA测定中,用含25%正常兔血清的PBS稀释血清与酶结合物可减少非特异性反应。采用抗原一次稀释包板供多次试验及多头移液器加样可减少试验的误差,提高反应的重复性。
English Abstract:
      High titer-effective soluble antigen was obtained after sonicating and high speed low tempreture (refrigerated) treatment of centrifuged monkey blood. The antigen absorbed on polystyrene plates could keep the antigenicity for at least twelve months at room tempreture. This method could be used for field detection and epidemiological survey. Non-specific reaction could be lowered through diluting sera and enzyme conjugate by 25% normal rabbit serum-PBS. Test errors might be decreased and the reproducibility enhaneed by using a batch of plates coated by diluted a antigen in a series of tests and by adding samples with a polymicropipette.
