Abstract:

目的探讨不同型别人乳头状瘤病毒(HPV)感染和端粒酶活化与宫颈癌的关系。方法对83例宫颈癌、47例宫颈上皮内瘤样病变(CIN)及10例正常宫颈应用TRAPPCR银染定性及TRAPPCRELISA定量法检测端粒酶活性,PCR法检测HPV分型。结果(1)端粒酶阳性率和HPV1618型阳性率:宫颈癌>宫颈CIN>正常宫颈,CINⅢ级>CINⅠ、Ⅱ级;HPV611型阳性率:CINⅠ、Ⅱ级>CINⅢ级。(2)宫颈癌中端粒酶阳性:Ⅰ期<Ⅱ期<Ⅲ、Ⅳ期;中分化>高分化;腺癌与鳞癌差异无显著性;HPV1618阳性:Ⅰ期<Ⅱ期<Ⅲ、Ⅳ期;HPV611阳性与临床分期、组织学分级及病理类型无关。(3)端粒酶阳性值:宫颈癌>CIN,CINⅢ级>CINⅠ、Ⅱ级;HPV611拷贝数:CIN>宫颈癌,CINⅠ、Ⅱ级>CINⅢ级;HPV1618拷贝数:宫颈癌>CIN,CINⅢ级>CINⅠ、Ⅱ级。结论HPV感染尤以1618型和端粒酶的激活与宫颈病变恶变关系密切

English Abstract:

Objective　To define a co rrelation between different human papillomavirus (HPV)types and telomerase activity in cervical cancer.Methods　Telomerase activity was detected by TRAP-PCR, and different HPV type was determined by PCR in 83 cervical cancer, 47 cervical intraepithelial neoplasia (CIN) and 10 normal cervix cases.Results With regard to positive rates of telomerase and HPV 16 18 :the results were cervical cancer >CIN >no rmal cervix, CIN Ⅲ >CIN Ⅰ, Ⅱ ;with regard to HPV 6 11 positive rate :the results showed CIN Ⅰ, Ⅱ >CIN Ⅲ.Positive rates of telomerase cervical cancer and HPV were bearing o n g rading and stag ing, but they did not correlate with histologic subtypes.Positive rate of HPV 6 11 had nothing to do with grading, staging and histolo gic patterns.On expression strength of telomerase and HPV 16 18:the results showed cervical cancer >CIN, CIN Ⅲ >CIN Ⅰ, Ⅱ.Reg ard to HPV 6 11'ex pression strength :the results showed CIN Ⅰ, Ⅱ >CIN Ⅲ, CIN > cervical carcinoma.ConclusionHPV 16 18 infection seemed to have played an important role in carcinog enesis of cervical lesions by activation of telomerase.
