目的 研究商品猪肝脏内戊型肝炎(戊肝)病毒(HEV)的检出状况及其基因分型。方法 采集35份山东省某个人屠宰场和某城镇大型屠宰场内的猪肝,处理后采用逆转录一巢式聚合酶链厦应(ncsted RT-PCR)方法检测HEV RNA。并与GenBank中登录的HEV全长序列进行同源性分析。结果 35份猪肝标本中共有3份检出HEV RNA,阳性率为8 57%。同源性分析显示这3株病毒均为Ⅳ型HEV。结论 即将上市的商品猪肝脏内存在HEv感染,其基因型与中国太陆HEV流行株相同,均为基因IV型。 |

Objective To determine the prevalence and genotype of hepatitis E virus (HEV) in swine liver before on the market and to analyze the phylogenetic relationship between the isolates from swine and human.Methods 35 swine liver specimens were collected from two slaughtering houses in the countryside of Shandong province,China.Nested reverse transcription-polymerise chain reaction (nested RT-PCR) and subsequent sequencing were used to determine the nucleotide sequences.A phylogenetic tree was constructed with Neighbor-joining method Eased on the Kimura-2-parameter model.Resutts 3 (8.57%)of the 35 swine liver specimens being texted were positive for HEV RNA.The three swine HEV strains isolated in the present study from liver samples shared the highest identity to genotype-IV HEV.Conelusion Results from the study confirmed that HEV was detectable among swine before on the market and the genotype was the same as that representing human and swine isolates in China.It also suggested that much more attention should be paid to the safety on the digestion of swine liver. |
