Abstract:
      目的寻找幽门螺杆菌(Hp)与胃癌相关蛋白。方法利用双向电泳分离Hp的全菌蛋白,应用ImageMaster2Dv3.1软件对3株分离自胃癌患者的Hp菌株、1株胃癌动物模型菌株及9株分离自非胃癌患者的Hp菌株的蛋白图谱进行比较,对目的蛋白进行基质辅助激光解析及电离飞行时间质谱分析,应用Mascot软件进行蛋白搜库。结果发现3种蛋白可能与胃癌相关,经肽指纹图谱鉴定,一种为酰基神经氨酸胞苷酰基转移酶,其Mowse分值为79,序列覆盖率为32%,另两种蛋白在现有的质谱库中无明确匹配蛋白存在。结论酰基神经氨酸胞苷酰基转移酶可能为与胃癌相关的Hp特异蛋白,另两种为新发现的可能与胃癌相关的Hp特异蛋白,其相关机制尚待进一步阐明。
English Abstract:
      Objective To identify specific; proteins of Helicobocter pylori(H. pylori) that associated with gastric carcinoma.Methods The whole-cell proteins of H.pylori were separated by two-dimensional electrophoresis (2-DE). The protein maps of four H. pylori strains associated with gastric carinoma anal nine strains that isolated from patients with non-gastric caroinoma were then compared by ImageMaster 2D v 3.1.MALDI-TOF mass spectrometry was performed to identlfy the proteins of interest. The proteins were searched by software mascot arid ldentlfled by peptlde flngerprlnt map.Results Three protelns Spemed to be associated wlth gastric carcinoma including acylneuraminate cytidylyltransferase with Mowse score 79 with the sequence coverage of 32 %. The other two had no unambiguous protein to match.Conclusions Acylneuraminate cytidylyltransferase seemed to be a specific H. pylori protein associated with the presence of gastric carcinnrna.Other two were novel proteins that might be associated with gastric carcinoma.However the mechanism needs to be explored.
