目的 探讨钩端螺旋体 (钩体 )毒力相关基因的分布特点。方法 根据钩体赖型赖株全基因组序列的生物信息学分析资料,选择了 12个可能与钩体毒力相关的基因,采用聚合酶链反应(PCR)方法,对中国问号钩体 38株参考菌株和 81株分离的野生菌株,以及 12株非致病性的双曲钩体共 131株菌株进行了检测。结果 问号钩体中各毒力相关基因分布广泛,双曲钩体中仅检测到个别毒力相关基因。lipL32基因存在于所有检测的问号钩体,lipL36基因在问号钩体不同菌株中的变异较大,阳性检测率为0 %~ 90.91% ;la1608基因在问号钩体黄疸出血群菌株中阳性检测率为87.5 0 %,而在其他血清群菌株间阳性检测率为0 %~ 25.00 %,sphA基因仅在问号钩体少数菌株中能检测到,阳性检测率为 17.65 %,且在中国赛罗群哈焦型参考菌株中未检测到。结论 这些基因可能是问号钩体重要的毒力相关基因。其中lipL32可能是问号钩体各血清群菌株共同抗原的编码基因 ;lipL36基因可能和问号钩体血清群特异性和多样性有关 ;la1608基因可能是黄疸出血群菌株特有的基因 ;哈焦型菌株与问号钩体菌株在基因结构具有较大的差异。 |

Objective To analyze factors related to the virulence associated genes of Leptospires.Methods Twelve putative virulence associated genes were detected by polymerase chain reaction(PCR) method in 38 reference strains, 81 field strains of Leptospira interrogans isolated from patients o r animals,and 12 avirulent strains of Leptospira bif lexa.Results These putative virulent genes were widely distributed among the strains of Leptospira interrogans, but only few of them w ere detected in Leptospirabi flexa.Gene lipL32 w as detected in all strains of Leptospira interrogans.Distribution of g ene lipL36 was varied sig nificantly with detected rates from 0 to 90.91 %.Gene la1608 had a po sitive rate o f 87.50 % fo rstrains of sero group Icterohaemorrhag iae, but was o nly detected in few strains of other serogroups with a range from 0 to 25.00%.Rate of detection on gene sphA was 17.65% in Leptospira interrogans, and was absent in serov ar hardjo reference strain.Conclusion Results indicated that these genes might be of importance for the virulence and pathog enicity of Leptospira interrogans, w hile gene lipL32 might be one of the common antig ens.Gene lipL36 might be involved in serogroup specificity w ith g enetic diversity, but genela1608 w as as one of the genes with specificity for serog roup Icterohaemorrhagiae.How ever, serovar hadjo might hold quite different genetic characteristics w hen compared with the other serov ars ofLeptospires. |
