Abstract: |
目的 利用免疫胶体金技术建立一种简便、快速并适用于基层人员使用的鼠疫菌抗原检测方法。方法 将抗鼠疫菌F1抗原的抗体致敏硝酸纤维素膜, 用于捕获鼠疫抗原, 然后用免疫胶体金颗粒进行标记。结果 应用鼠疫菌免疫胶体金快速检测法最低可检出不同鼠疫菌(EV和Evp株)1.56×105CFU/ml, 检测鼠疫菌F1抗原最低可检出1ng/ml, 与鼠疫反向间接血凝法的检测结果一致; 并且不与假结核小肠结肠炎等耶尔森菌以及其他相关细菌发生交叉反应。结论 鼠疫菌免疫胶体金快速检测法具有较好的特异性、敏感性和简便快速的特点。 |
English Abstract: |
Objective To develop a method of immunochromatography assay( ICA) with sensitive,
specific, rapid, simple and suitable for the detection of Yersinia pestis antigen at the local laboratories.
Methods Colloidal gold labeled with the anti-F1 antibody of Yersinia pestis, was connnected with the
anti-F1 antibody of Yer sinia pestis to pyroxylin membr ane and assembled them to the dipstick of ICA.
Results Results showed t hat the rates of sensitivity for F1 antig en and Yersinia pestis were 1 ng/ ml and
1. 56×105 CFU/ ml respectiv ely. However, Yersinia pseudotuberculosis et al could not be detected by
dipstick of ICA. Conclusion The method of ICA appeared to be consistent to those of r-IHA with better
specificity and sensitivity but was simple and rapid for the detection of Yersinia pestis and F1 antigen. |
