Abstract:
      目的研究湖南省啮齿动物中汉坦病毒(Hv)流行情况及病毒型别。方法采用IFA法检测鼠肺中HV抗原;用RT—PCR法扩增阳性标本中Hv的部分s和M片段;构建系统发生树进行系统发生分析及分型。结果在湖南省HV流行的地区共捕获啮齿动物344只,其中6份标本HV抗原阳性,病毒携带率为1.74%。对扩增出的部分S与M片段的核苷酸序列分析表明,5份标本中的病毒为汉城型HV(SEoV),l份为汉滩型HV(HTNV)。用S(620~990nt)与M片段G2区(200l~2301nt)核苷酸序列所构建的系统进化树显示,湘乡褐家鼠、黄胸鼠及黄毛鼠携带的病毒均为sEOV的S4亚型;宁远褐家鼠携带的病毒为sEOV的一个新亚型;石门小家鼠携带的病毒为HTNV的H4亚型。结论湖南省为混合型HV疫区,以汉城病毒为主,并具有宿主多样性。
English Abstract:
      Objective To explore the hantavirus infection and their genotype in rodents in Hunan.Methods Hantavirus antigens in the rat lungs from Hunan province were detected by immunofluorescenceassay. Partial S and M segment in antigen-positive samples were amplified by RT-PCR, and then sequenced.The phyologenetic trees were constructed for the analysis of genetic characters of hantavirus. Results Atotal of 344 rats were trapped in the main epidemic area of Hunan province,and hantavirus antigens werefound in 6 of the 344 rats(1.74%).The phylogenetic trees constructed by partial S segment( nt 620-990 )or partial G2 segment(nt 2001一2301)showed that the hantaviruses carried by Rattus norvegicus,R.fZabipectus and R. rattoides from Xiangxiang district were genetic subtype SEOV4. The virus carried byR.norvegicus in Ningyuan district was phylogenetically different from the known SEOV. The hantaviruscarried by Mus musculus from 5himen district was genetic subtype HTNV4. Conclusion The hantavirusesin the main epidemic areas in Hunan province mainly belonged to SEOV, and R.flabipectus andR.rattoides carried the same genotype of SEOV as R.norvegicus.
