Abstract: |
目的调查黑龙江省东部中俄边境口岸蜱的种类、分布、携带莱姆病螺旋体情况及病原基因型。方法捕蜱采用布旗法;蜱带莱姆病螺旋体率采用直接荧光法检查;病原体分离培养采用BsKⅡ培养基,对分离到的螺旋体用单克隆抗体(H5。、地。,。、H9,:。)、sD$PA(沲蛋白图谱分析、PCR—RFLP分析以及基因测序等方法进行表型和基因分型研究。结果黑龙江省东部7个中俄边境口岸蜱的种类为1科3属4种,即硬蜱科硬蜱属全沟硬蜱,革蜱属森林革蜱,血蜱属嗜群血蜱和日本血蜱。不同生态环境中蜱的分布不同,各口岸优势蜱种不同。全沟硬蜱带莱姆病螺旋体率较高,平均为31.4%,森林革蜱和嗜群血蜱的带菌率较低,平均为2.2%和3.8%,日本血蜱中未检出阳性者。首次在中俄边境口岸——东宁、同江采集的全沟硬蜱中分离到伯氏疏螺旋体并鉴定为B.舭r而ii基因型。结论黑龙江省东部边境口岸为莱姆病的自然疫源地。 |
English Abstract: |
Objective To explore the fact that the east border of Heilongjiang province had been alyme disease natural focus, we investigated the species and distribution of ticks and isolated bacteria fromticks and identified genomic species of Borrelia burdorferi sensu lato. This study provided evidence forprevention and control of Iyme disease. Methods Ticks were caught by flagging method and Directimmunofluorescence method was used to detect the rate of bacteria borne by the tick. BSK Ⅱ culturemedium was used to isolate the agent and印ecific McAbs were used to identify the bacteria. SDS-PAGEprotein profile and PCR-RFLP method were also used to identify the species of Spirochetes. Results Ticks,collected from China-Russia border of east Heilongjiang province were classified including Ixodes persulcatusSchulze, Dermacentor sivarum Olener, Haemaphysalis concinna Kock,and Haema灿ysalis japonica Kock.We found that the distributon of ticks was different under different circumstances and the predominantspecies were also different in different ports. The rate of bacteria borne by Ixodes persulaatus Schulze was31.4 %, by Dermacenror sivarum Olener and Haemaphysalis concinna Kock were 2.296 and 3.8 96,respectively. However,it was negative for Haemaphysalis japonica Kock. Spirochetes isolated from Ixalespersulcatus Schulae were collected from Dongning and Tongjiang while Genomic species of Spirochetes,isolated from ticks of the border belonged to B. garinii. Conclusion All the Results showed that the eastborder of Heilongjiang province was the natural focus of lyme disease. |
