Abstract:
      目的<\b>对2004年深圳市一起肠道病毒71型(Ev71)引发手足口病流行的基因型别分析。方法<\b>用EV71型特异性引物进行RT.PCR,并对EV71的VPl和VP4基因进行克隆,所得的序列与EV71型A、B、C基因型代表株的核苷酸序列用TreeView和PHYLIP软件(3.6b)进行系统进化分析。结果<\b>4株病毒与C基凶型代表株比较接近。VPl区核苛酸同源性在87.8%~92.0%之间,VP4区核苷酸同源性在85.9%--87.4%之间;与A、B基因型代表株比较差异较大,VPI区核苷酸同源性为81.9%--84.2%,VP4区为80.6%~85.0%;4株病毒VPl核苷酸同源性为94.1%~99.8%,vP4同源性为100%,组成一个独立的小分支。结论<\b>对EV71的VPI和VP4区进行基因进化分析,可得出类似的结果<\b>,4株Ev7l深圳流行株可命名为CA亚型。
English Abstract:
      bjective To carry OUt genotype analysis of enterovirus type 71,detected from hand— foot-mouth disease patients in Shenzhen in 2004.Methods AIl samples were tested by reverse transcription—polymerase chain reaction(RT-PCR)using EV71 specific primer.The VPl and VP4 of EV71 were cloned and sequented.A phylogenetic tree was constructed by comparing the sequences with genogroups A,B and C genotypes using TreeView and PHYLIP software(3.6b).Results The VPl nucleotide sequence of 4 strains iselated from Shenzhen shared 87.8%一92.O%identitywith genogroup C,while the homogeneity of the VP4 nucleotide sequence was between 85.9%一87.4%.The homogeneity of the VPl nucleotide sequence with genotypes A and B WflS between 81.9%一84.2%and WaS 80.6%.85.0% with VP4.Among the 4 strains.the homogeneity of the VPl nucleotide sequence Was between 94.1%. 99.8%and was 100%with VP4 which formed asinail group and could denominate EV7l genetype CA.Conclusion Similar results were obtained from phylogenetic analysis of EV71 based on VPl and VP4 nucleotide sequence.The four EV71 strains causing epidemic in Shenzhen could be named硝CA subgroups.
