Abstract:
      目的对2008年河南省手足口病患者体内分离的肠道病毒71型(Ev71)进行全基冈组核苷酸序列测定。方法采用RT-PCR扩增覆盖全基因组的8个重叠基因片段,双向测序,DNAStar软件中的MegAlign等软件进行同源性分析,MEGA3软件进行进化树分析和做图。结果8个重叠基因片段序列拼接得到EV71全基冈组序列,共7405个核苷酸,与其他EV71病毒基因组构成相似,在非编码区有核苷酸的插入和缺失。同源性分析:在P1区,HENAN08株与安徽株(AnhuiFY08)、深圳株(SHZH98和SHZH03)、浙江株(Zhejian908)等毒株的核苷酸和氨基酸同源性分别大于91%和97%;与标准株(BrCr)和台湾流行株(TW2086)的核苷酸和氨基酸同源性分别大于75%和82%;与CoxsackievirusAl6(Cox.A16)的同源性最低,为68.7%和79.6%。在P2和P3区,HENAN08株与其他国内EV71株的同源性均较高,而与BrCr以及TW2086的同源性低于Cox.A16。Pi区遗传进化分析表明:HENAN08株与AnhuiFY08和Zhejian908的进化关系都密切,与Cox.A16疏远。结论HENAN08属EV71病毒的C4亚型,与AnhuiFY08、Zhejian908和SHZH03进化途径类同。
English Abstract:
      objective To sequence the whole-genome of enterovirus 71(EV71)srtainisolated from patient with hand.foot and mouth in Henan province in 2008.Methods Eightoverlapping clones covering the whole viral genome were obrained by RT-PCR and the sequenceswere determined by Sanger dideoxg.mediated chain termination method.Results Data it showed that the full length of enterovirus 7 l(EV71)HENAN08 genome(not including Poly Atail)iS 7405 bp.NO deletion or insertion was detected in the coding region.There were several deletions and insertions in 5’UTR and 3’UTR regions.In Pl region.HENAN08 strain shared high homology with AnhuiFY08 strain,Zhejian908 strain and SHZH strains(SHZH98.SHzH03)but lOW homology with COX.A16.In P2 and P3 regions.HENAN08 strain shared higher nucleotide homology with COX.A16(81.7%and 83.7%)than that with BrCr and TW2086 strains.The phylogenetic analysis based on PI region demonstrates that HENAN08 strain had the nearest genetic relationship with AnhuiFY and Zhejiang strains(isolated in 2008).Conclusion The HENAN08 strain might belong to the same genogroup with AnhuiFY08 and Zhejian908 strains as C4 gene subtypes.
