Abstract:
      目的了解HIV-!毒株亚型在深圳地区吸毒人群中的流行情况,并分析其流行趋势及进化规律.方法收集1996~2008年深圳地区吸毒人群HIV确认阳性样本166例,应用巢式聚合酶链反应(nPCR)技术,对样本膜蛋白基因(env)进行扩增,并对其基因区核苷酸序列进行测定和分析.结果166份样本中共存在CRF01-AE、CRF07- BC、CRF08-BC 3种重组毒株以及B'、C、A1 3种亚型|其在所有分析样本中的比例分别为65.06%(108/166)、19.88%(33/166)、6.02%(10/166)、 7.23%(12/166)、0.60% (1/166)和1.20%(2/166)|系统进化分析发现,不同时间段HIV-1各亚型出现较明显的汇聚现象|3个不同时间段所流行的CRF01 AE、CRF07 BC亚型组内及与标准参考株间的基因离散率有较为显著性的增大趋势.结论深圳地区吸毒人群HIV-1流行毒株以CRF01 AE重组亚型为主,其次是CRF07 BC重组亚型,也存在CRF08 BC、B'、 C、AI亚型,并且各亚型在流行过程中已发生较大变异.
English Abstract:
      Objective To study the prevalent status of human immunodeficiency virus-l(HIV_1)subtypes in IDU(injecting drug users)population in Shenzhen and to study their source of infection in order to predict the epidemic trend and evolution.Methods166 HIV-1 positive plasma from the IDUs was collected from 1 996 to 2008.HIV-l env genes were amplified by nested-PCR from RNA.The C2-V3 regions (450 bp)of HIV-1 env were sequenced for analyses.Phylogenetic analyses were performed on the nucleotide sequence data. Results Among 166 samples.there were 6 HIV-l strains including CRF01_AE.CRF08_BC, CRF07_BC 3 circulating recombinant forms (CRFs)and B', C, A1 3 subtypes.Data from the genotype analyses showed that 65.06%(108/166)were CI0l AE, 19.88%(33/166)were CRF07_BC.6.02%(10/166)were CRF08_BC, 7.23%(9/166)were subtype B'.O.60%(1/166)were subtype C and 1.20%(2/166)were subtype AI.Phylogenetic tree analysis showed that some of HIV-1 clusters defned in CRF01_ AE, CRF07_BC and subtype B'in different time groups.Significant increase of gene distance in CRF0l-AE and CRF07-BC strains in the three different periods.Conclusion CRF0l_AE and CRF07_BC were the major epidemic CRF strains among the IDU population in Shenzhen while the subtype B', C, A l and CRF08_BC were also circulating in IDU population in this region.The variation of all different subtypes was increasing through these years.
