Abstract: |
目的运用分子生物学方法对水痘或带状疱疹患者皮肤水疱液及水痘疫苗中分离得到的水痘一带状疱疹病毒(VZV)株进行基因特征分析。方法从水痘疫苗、水痘或带状疱疹患者的皮肤水疱液中分离VZV,然后利用聚合酶链反应和限制性片段长度多态性对病毒株的ORFs6、38、62进行分析,同时测序分析ORFs 1、3l、51和62中的单个核苷酸多态性位点。结果4种疫苗酶切结果均为Alu I-Pst I-Sma I+BssHI+Nae I+,49株ⅥⅣ临床分离株巾Alu I+Pst I+Sma I BssH 1I-Nae I-占96%(47株),Alu I-PSTI-Sma I+BssHlI+占2%(1株),Alu I+Pst I-Sma I-BssHII NaeI占2%(1株),其中l例患者的临床分离病毒株与疫苗一致;测序分析显示不同疫苗核苷酸序列之间存在差异,离得到的疫苗株病毒序列与Varilrix一致。结论存在疫苗引起水痘的情况,测定ORFs 6、62中的限制性片段长度多态性能够区分疫苗株和野毒株,对ORFs l、3l、5l和62中的单个核苷酸多态性位点进行分析可追踪疫苗致病的病毒株来源。 |

Objective To study the genetic characteristriCS of vaccine and variceila-zoster virus(VZV)strains isolated from patients wi山chickenpox or zoster by molecular analysis.Methods SNP based VZV genetic charaetefistrics were analyzed in 19 VZV isolates using the
restriction fragment length polymorphisms analysis of DNA fragments of the open reading frames 6,38,62 and sequence alignment of the opell reading frames l。3 l,5 l,62.Results A11 vaccine strains wererevealedAlu I-Pst I-Sma I+BssH1I+Nae I+,96%clinicalisolateswere revealed Alu I+Pst I+Sma IBssHlI-Nae I-,2%clinical isolates were revealcd Alu I-Pst I-Sma I+BssHⅡ+,2%clinical isolates WCl"e revealed Alu I+Pst I-Sma I-BssH II Nae I’bv restriction fragment length polymorphisms analysis and sequence alignment revealed the mutations also presented in this four vaccine strains.Conclusion Use the restriction fragment length polymorphisms analysis of DNA fragments of the open reading,frames 6 and 62 could be distinguished VZV wild-type strains and vaccine strain in clinical isolatcs in China.In order to fmd the adverse effect caused by vaccine from certain company’S,analysis on the SNPS in ORFs 1,3l,51 and 62 is needed. |
