| Abstract: |
| 柯萨奇病毒A6(CVA6)属于肠道病毒A组成员，主要引起疱疹性咽峡炎，也可引起手足口病.2011年河南地区从手足口病患者粪便标本中分离到1株CVA6，为了解其基因特征，本研究对分离的CVA6毒株进行全基因序列测定.1.材料与方法:待测毒株为2011年河南省疾病预防控制中心传染病预防控制所实验室分离并鉴定的毒株，命名HN421.引物设计:根据原型株Gdula(AY421764)和日本毒株Shizuoka 18 (AB678778)序列，设计10对首尾相互重叠、覆盖全基因的引物(表1).RNA提取及反转录:将毒株接种于RD细胞，待病变达到75％时收获毒株，用美国Qiagen公司的Rneasy mini试剂盒提取RNA，用大连宝生物公司的PrimeScript 1-Strand cDNA Synthesis试剂盒反转录cDNA，所有操作均按说明书进行. |
